Геном тұрақтылығы жасушалардың тіршілікке қабілеттілігінің, организмдердің эволюциялық тұрақтылығының және олардың функционалдық тұтастығын сақтаудың негізі болып табылады. Эндогенді зақымданулар, әсіресе жасушалық метаболизм нәтижесінде пайда болатын азотты негіздердің тотығу арқылы модификациялануы, ДНҚ құрылымын бұзатын негізгі факторлардың бірі болып саналады. Мутациялардың жиналуын болдырмау үшін жасушалар негіздерді кесіп алып тастау арқылы жүретін репарация жүйесін (BER) қолданады, мұнда ДНҚгликозилазалар шешуші рөл атқарады. Бұл ферменттер зақымданған немесе қате жұптасқан негіздерді танып, оларды алып тастау арқылы репарация реакцияларының каскадын бастайды.
MutY/MUTYH тұқымдасына жататын адениндік ДНҚ-гликозилазалар зақымданған негіздерді тікелей алып тастамай, гуаниннің тотығу зақымдануы нәтижесінде туындайтын репликациялық қателерді түзетуімен ерекшеленеді. Бактерияларда бұл қызметті MutY ферменті атқарса, эукариоттарда оның гомологы – MUTYH орындайды. Бұл ферменттер G:C → T:A типті транзициялық мутациялардың бекітілуінің алдын алып, спонтанды мутагенез деңгейін едәуір төмендетеді.
